I went into Control Panel's "Password Protect Directories" &  protected the subdirectory & I added Active Users/passwords. Now when view in "Password Protect Directories", the subdirectory has the padlock locked icon.

However, receive Error 403 Forbidden when attempt to access a file within the protected directory via web.  The .htaccess file located within the protected directory = 640.  

I want employees to be able to access files in the protected directory with an ID & Password & just have to Login once for each session.   TIA

Usernames and passwords are case sensitive.  Try adding another user and authrorizing.  If it still doesn't work, send an email to support.

Installing or removing your FrontPage extensions will disable directory protection, and it would have to be redone.

If you do use FrontPage, you can also use that to protect directories.
Click Tools... Server...Administration and make a subweb.  Then you can set up users for that subweb (a directiry in your website).
